Collect blood by into a pre-chilled 6 ml vacutainer (lavender top) containing EDTA as the anti-coagulant. Immediately add 300 ul of the ready-to-use aprotinin solution (Sigma #A6279). Mix by inversion and place sample in an ice bath immediately. Centrifuge the blood within 10 min of collection at 1500 x g for 15 minutes in a refrigerated centrifuge set at 4 C◦. Transfer the clear plasma to a labeled 12 x 75 mm polypropylene tube. Store at -80◦ until time of assay.
The Cayman Chemical Company enzyme immunoassay (EIA) requires 2 ml of plasma which is extracted and concentrated prior to assay.
